In general, yes. There is no well-established evidence that these three ingredient categories are inherently incompatible when used in properly formulated topical skincare.
Their proposed roles also differ. Niacinamide is commonly used for barrier support and visible tone; peptides are chosen for peptide-specific signalling or conditioning roles; ectoin is used primarily for hydration, stress-protection mechanisms and barrier comfort.
The more important question is not whether they can coexist, but whether the finished product is stable, well tolerated and formulated at meaningful levels.

Niacinamide is one of the more extensively studied cosmetic ingredients. Clinical studies have reported improvements in fine lines, hyperpigmented spots, texture, elasticity and other visible facial-skin endpoints in tested formulations.
It has also been associated with improved barrier-related measures such as reduced transepidermal water loss in some studies.
Its role at night is therefore broader than 'brightening': it can be positioned more accurately as a multifunctional skin-support ingredient.

Peptides are short chains of amino acids, but cosmetic peptides are not interchangeable. Different sequences are designed for different roles, and evidence for one peptide cannot automatically be transferred to another.
Clinical studies of certain peptide-containing regimens have reported improvements in wrinkle appearance and skin smoothness. However, many such products also contain niacinamide, retinoid derivatives, antioxidants or moisturising systems.
It is therefore too strong to say that 'peptides rebuild collagen overnight'. A more defensible statement is that selected cosmetic peptides may support the appearance of smoother or firmer-looking skin when used in appropriately formulated products.

Ectoin is an extremolyte originally studied in microorganisms that survive harsh environmental conditions. In topical skincare, it is valued for its strong interaction with water and its use in formulations designed to support skin comfort and barrier function.
A randomized vehicle-controlled trial of a 2% ectoin formulation reported improvements in moisturisation, skin surface structure and elasticity compared with control conditions.
Other ectoin-containing formulations have also been studied in barrier-compromised skin, although results from those populations should not be extrapolated directly to healthy cosmetic users.

Niacinamide, peptides and ectoin overlap at the broad level of supporting healthier-looking skin, but their proposed mechanisms are not identical.
Niacinamide has evidence across barrier function and visible tone. Ectoin is particularly relevant to hydration and stress-related skin comfort. Peptides may contribute peptide-specific signalling or conditioning benefits.
That makes the combination conceptually complementary. However, complementary mechanisms do not automatically establish additive or synergistic clinical effects.

None of these three ingredients requires darkness in order to function. Their inclusion in nighttime skincare is primarily about routine design, skin comfort and pairing with an overnight moisturising environment.
Night is also a practical period for applying leave-on products without immediate exposure to sweat, makeup or repeated cleansing.
It is therefore more accurate to describe the combination as well suited to an overnight routine than to claim that the ingredients are biologically active only at night.

That depends on skin type, climate and the complete formula. A traditional night cream often relies on emollients and occlusives to reduce water loss and create a richer moisturising film.
A lightweight multi-active product can provide hydration and targeted ingredient support, but very dry or barrier-compromised skin may still prefer or require a richer moisturiser.
The scientifically responsible distinction is between format and function: a mist, serum or fluid can deliver actives, but it does not automatically replicate the occlusivity of a cream.

Current evidence supports niacinamide as a multifunctional topical active, ectoin as a hydration- and barrier-support ingredient, and selected peptides as potentially useful cosmetic actives when evaluated in their specific formulations.
Using the three together is scientifically reasonable because their roles are not inherently incompatible or identical.
What the evidence does not establish is that the trio is automatically synergistic, superior to other combinations, or clinically proven in every finished product.
